6. WFA staining results showed that the area of PNNs in the hippocampal region of mice in the EtOH group was significantly reduced (P < 0.001, n = 4). Overexpression of ICAM5 led to a significant increase in the area of PNNs (P < 0.01, n = 4), with the most pronounced changes observed in the CA1 region.
7. Phalloidin staining results indicated that F-actin in the CA1 region of the hippocampus in the EtOH group was significantly increased (P < 0.01, n = 4). Overexpression of ICAM5 resulted in a significant decrease in F-actin (P < 0.001, n = 4).

Analysis of the expression levels of cytoskeletal proteins F-actin and G-actin: Western blot results showed that compared to the Water group, the ratio of F-actin to G-actin in the hippocampal region of mice in the EtOH group was significantly increased (P < 0.05, n = 5). Overexpression of ICAM5, i.e., in the ICAM5 + EtOH group, led to a significant decrease in the F-actin/G-actin ratio compared to the EtOH group (P < 0.05, n = 5).
9. Detection of the expression levels of cytoskeleton-related proteins CaMKII and Arc showed that chronic alcohol exposure significantly increased the level of CaMKII autophosphorylation (P < 0.05, n = 5) and the expression level of Arc (P < 0.0001, n = 6). Overexpression of ICAM5 led to a significant decrease in the level of CaMKII autophosphorylation (P < 0.05, n = 5) and the expression level of Arc (P < 0.01, n = 6).
