To investigate the expression status of the DKK1 protein in clinical cancer tissues, we stained the tissue sections using a rabbit polyclonal antibody specific to human DKK1 (Santa Cruz) raised against the NH2-terminal portion (amino acids 1–120) of DKK1, as previously described (31). The specificity of the anti-DKK1 antibody was confirmed by Western blot analysis using lysates of DKK1-expressing NSCLC and ESCC cells (31), and those that were transfected with small interfering RNA duplexes against DKK1 or control small interfering RNAs (Supplementary Fig. S1A and B).
